Abstract:

三联吡啶钌电化学发光淬灭生物传感器的制备方法,属于生物传感器制备领域。包括步骤:1)确定待检测miRNA,并遵循序列互补原则设计发卡探针;2)在玻碳电极表面电沉积氧化石墨烯;3)1‑芘丁酸‑N‑羟基琥珀酰亚胺酯(PASE)与亚甲基蓝(MB)同时竞争吸附在电还原氧化石墨烯表面;4)3‑叠氮基‑1‑正丙胺(3‑Azido‑1‑PrA)与PASE发生酰胺反应,将电极表面叠氮化;5)合成Ru(bpy)2(dcbpy)NHS标记的电化学发光探针Ru‑DNA;6)Ru‑DNA发生点击化学反应组装到电极表面。7)当目标miRNA存在时,阻碍发光物质Ru(bpy)2(dcbpy)NHS与淬灭剂MB之间共振能量转移的发生,引起电化学发光信号强度改变。本发明可实现对目标miRNA的灵敏、快速、简便检测。
