Abstract:

增强鲁米诺体系的电化学发光生物传感器的制备方法,属于生物传感器制备领域。本发明步骤:1)确定待检测的目标miRNA,并根据序列遵循碱基互补原则设计DNA探针序列;2)通过电化学循环伏安法,在玻碳电极表面电沉积金纳米颗粒,并将设计的DNA探针(5’端修饰巯基)通过金硫键连接到金纳米颗粒上;3)使用葡萄糖氧化酶封闭电极表面的非特异性结合位点;4)将目标miRNA与封闭后的电极反应,与DNA探针杂交后形成双链复合物;5)在杂交后的电极上滴加血红素溶液,该物质只嵌入miRNA‑DNA双链之间,可有效加速过氧化氢的分解并生成超氧阴离子,从机理本质上实现电化学发光的增强。本发明解决了鲁米诺体系发光信号弱,生物传感器检测灵敏度低的问题。
