Abstract：
一种利用硅油的适用于微滴数字PCR的微滴制备方法，配制PCR水相，将上游引物、下游引物、PCR模板、PCRmix依次均匀混合，滴加双蒸水使溶液体系并分装3个管，各管体积相同并作为反应水相备用；配制PCR油相，取二甲基硅氧烷作为反应油相备用；取水相PCR加到油相中，涡旋震荡使油水两相充分乳化，得到油包水微滴；本发明提供的制备方法，可便捷地配制一种具有颗粒均匀、热力学性能稳定的油包水反应微滴，简便了微滴数字PCR实验的操作。
