Abstract:

本发明公开了一种氨氧化古菌的实时荧光定量PCR检测方法,包括以下步骤:步骤1,引物的合成,其中,引物的序列为:正向引物AOA_amoA_F:5'-TTCTACACAGACTGGGCGTG-3';反向引物AOA_amoA_R:5'-GTTAGGTCCAAAAGCATCGC-3';步骤2,普通PCR、克隆与测序鉴定引物的特异性;步骤3,标准品质粒的制备;步骤4,实时荧光定量PCR反应体系和反应程序的建立;步骤5,标准曲线的绘制:步骤6,实时荧光定量PCR检测方法的评价。本发明的有益效果为:实时荧光定量PCR引物的特异性强,实时荧光定量PCR的检测方法具有灵敏度高、重复性及稳定性和准确性好,简便快速,可同时实现对于不同环境样品中氨氧化古菌的定量,对于研究氮循环及环境样品中的氨氧化过程具有重要意义。
