Abstract:

一种兼有抗耐药性、生物素受体靶向性和低氧/低pH双重响应性的多功能纳米载体及其制备方法和应用属于生物医用高分子材料技术领域。本发明提供的生物素受体靶向和低氧与低pH双响应的AGT抑制剂偶联物,包括疏水性的2‑氨基‑O<supgt;4</supgt;‑苄基蝶啶基团(BD)、低氧响应基团偶氮苯(AZO)和生物素(Bio)修饰的亲水性高分子材料。该体系可通过分子间作用力将疏水性化疗药物装载于载药体系中,靶向肿瘤细胞上的生物素受体,并利用其高渗透性和长效性将其富集于肿瘤组织;以低氧和低pH为特征的肿瘤微环境可诱导纳米载体上偶氮键断裂及pH响应,释放出具有抑制AGT活性的BD和具有抗肿瘤活性的烷化剂,从而提高药物的抗耐药性和抗肿瘤活性,并增加了靶向性。
