Abstract：
大肠杆菌中具有产氢功能基因的载体pET32a‑fdhF‑1及其构建和应用，涉及基因工程技术领域。pET32a‑fdhF‑1基因载体为包含有大肠杆菌BL21甲酸脱氢酶fdhF基因的pET32a质粒。其构建是通过设计引物扩增大肠杆菌BL21的甲酸脱氢酶(formate dehydrogenase H)的fdhF基因片段，并通过T4 DNA连接酶将其连接到酶切后的pET32a载体中，命名为pET32a‑fdhF‑1，进行基因测序，验证基因的完整性。将pET32a‑fdhF‑1质粒转化到大肠杆菌中，在产氢培养基中进行培养产氢。转入载体pET32a‑fdhF‑1能提高大肠杆菌产氢效率。
