Abstract:

一种针对膨胀污泥中H.hydrossis丝状菌群聚合酶链式反应方法,涉及污水生物处理领域。H.hydrossis丝状菌群聚合酶链式(PCR)方法反应体系:2倍浓度的Taq PreMix试剂10μl;DNA模板0.5‑1μl;正向引物;反向引物;加灭菌水到总体积20μl。反应程序:①1个循环:温度为94℃时间为2‑5min。②30个循环:依次为变性温度94℃,时间20‑30s;复性温度50‑55℃,时间30s‑1min;延伸温度72℃,时间2min。③1个循环:温度72℃,时间5‑10min。PCR反应后将扩增产物在1.5%的琼脂糖凝胶电泳中跑胶,得到分离良好的DNA亮条带。本发明可以高效PCR反应扩增,对H.hydrossis丝状菌群进行鉴定分析,为后续该菌种相关菌群发育树分析及菌群绝对定量分析提供基础。
