Abstract:

高效降解油脂功能基因载体及其构建和应用涉及基因工程技术领域。基因载体pMAL‑p5X‑lipase为包含有蜡状芽孢杆菌脂肪酶基因lipase的pMAL质粒。其构建过程是通过设计引物扩增蜡状芽孢杆菌中脂肪酶基因lipase片段,并用重组酶将其连接到malE基因下游,进行基因测序,验证基因的完整性。将pMAL‑p5X‑lipase质粒转化到细菌中,在加入油的试管中测定此细菌降解脂肪能力。pMAL载体用于pMAL蛋白融合表达和纯化系统,含有Ptac强启动子、MBP起始翻译信号及多克隆位点,能够将目的基因与malE基因(编码MBP蛋白)融合表达。pMAL‑p5X载体含有malE信号肽序列,它将引导融合蛋白穿越质膜,能够以大肠杆菌为宿主菌进行基因表达。
