Abstract:

本发明涉及一种基于适配体靶向递送microRNA的纳米载体及其制备方法与应用。其中靶向分子为巯基修饰的MUC-1分子,可与抑癌性microRNA-34a结合,使MicroRNA-纳米金刚石-鱼精蛋白复合体(MNPs)成为具有主动靶向功能的AMNPs。通过静电相互作用,将带负电荷的aptamer-microRNA(Apt-miR)嵌合体吸附于阳离子性鱼精蛋白-纳米金刚石基载体,构成三元复合载体。进行纳米转运体系对非小细胞肺癌的治疗效果的研究。Apt-miR嵌合体免疫原性小,经修饰的MUC-1核酸适配体在生物体内化学稳定性好,并且易于化学合成。为提高其转染效率,使用鱼精蛋白修饰的纳米金刚石材料(NPs)帮助miRNA通过渗透和滞留增强效应增加其在肿瘤部位的积累。
