Abstract:

According to the growth and metabolism characteristics of CM1 cell, a new low-serum medium was successfully developed, which was added 2% new calf serum (NCS) in the base of medium. In comparison with the culture in the medium with 10% fetal bovine serum (FBS), the morphology was the same and the growth was better in the culture with 2% NCS. A maximum cell density, 24.2105/mL, was obtained at 120 h in the culture with 4 mmol/L glutamine, and increased by 80.6% than that in the culture without glutamine. The number of cells in S phase decreased with the increase of initial glutamine concentration, and it was benefit for the synthesis of DNA. With the increase of initial glutamine concentration from 0 mmol/L to 4.0 mmol/L, The specific consumption rates of glucose decreased by 16.5%, and the consumption of cell to lysine, valine, and isoleucine decreased significantly. The key metabolic enzymes activities of hexokinase, pyruvate kinase, and lactate dehydrogenase decreased with the increase of glutamine, showing that the flux of glycolysis was decreased. A higher activity of glutamate-oxalacetic aminotransferase showed that the transamination pathway was still capital in the metabolism of glutamine.
